Adefovir dipivoxil is a prodrug of adefovir, an acyclic nucleotide analog of adenosine monophosphate. It is phosphorylated intracellularly to adefovir diphosphate, which inhibits hepatitis B virus (HBV) DNA polymerase by competing with the natural substrate deoxyadenosine triphosphate and causing DNA chain termination after incorporation into viral DNA.
Inhibits influenza A virus uncoating and viral RNA replication; increases dopamine release and blocks dopamine reuptake in the CNS.
10 mg orally once daily on an empty stomach.
100 mg orally twice daily; may increase to 200 mg orally twice daily if tolerated, usually in divided doses. For Parkinson's disease, 100 mg orally twice daily; for drug-induced extrapyramidal reactions, 100 mg orally twice daily.
