Mechanism of Action
AZASITE

Azasite (azithromycin ophthalmic solution) is a macrolide antibiotic that binds to the 50S ribosomal subunit of susceptible bacteria, inhibiting protein synthesis.

DIASTAT

Diazepam enhances the effect of gamma-aminobutyric acid (GABA) at GABA-A receptors, increasing chloride ion conductance and neuronal hyperpolarization, leading to anxiolytic, sedative, muscle relaxant, and anticonvulsant effects.

Treatment of bacterial conjunctivitis caused by susceptible organisms

AZASITE

Azasite (azithromycin ophthalmic) is classified as FDA Pregnancy Category B. Systemic absorption is minimal after ophthalmic administration. No teratogenic effects have been observed in animal studies at doses up to 200 mg/kg/day (systemic). Limited human data; risk is considered low. First trimester: unlikely to cause major malformations. Second and third trimesters: no specific risks identified.

DIASTAT

DIASTAT (diazepam) is classified as Pregnancy Category D. First trimester: Increased risk of congenital malformations, particularly cleft lip and palate, when used during the first trimester. Second and third trimesters: Chronic use may lead to fetal dependence and withdrawal symptoms postnatally; risk of floppy infant syndrome (hypotonia, lethargy, sucking difficulties) when administered near term.

Lactation Summary
AZASITE

Azithromycin is excreted into human milk after systemic administration; the M/P ratio is approximately 0.90. Considered compatible with breastfeeding; use with caution if eye drops are applied multiple times daily.
