BETAPRONE (propiolactone) is an alkylating agent that exerts its effects by cross-linking DNA and RNA, leading to inhibition of cellular replication and transcription. It also acts as a chemical sterilant by inactivating proteins and nucleic acids through covalent modification.
Halobetasol propionate is a high-potency corticosteroid that exerts anti-inflammatory, antipruritic, and vasoconstrictive effects via binding to glucocorticoid receptors and modulating gene expression. Tazarotene is a retinoid prodrug that is converted to its active metabolite, tazarotenic acid, which binds to retinoic acid receptors (RAR-β, RAR-γ) to regulate gene expression involved in cell proliferation and differentiation.
Not established; BETAPRONE is an experimental agent with no approved dosing. In clinical trials, doses of 0.5-2 mg/m² IV weekly have been used.

For halobetasol propionate, renal excretion of metabolites accounts for ~80% and fecal ~20%. For tazarotene, renal excretion of metabolites accounts for ~60% and fecal ~40% after oral administration, but topical absorption is <1%.
