Danazol is a synthetic androgen derived from ethisterone. It suppresses the pituitary-ovarian axis by inhibiting gonadotropin (LH and FSH) release, leading to anovulation and amenorrhea. It also binds to androgen, progesterone, and glucocorticoid receptors, exerting weak androgenic, antiestrogenic, and antigonadotropic effects. Additionally, it may directly inhibit ovarian steroidogenesis and increase clearance of endogenous sex hormones.
Testosterone replacement therapy; testosterone binds to androgen receptors, activating gene transcription for male sexual development and maintenance of secondary sexual characteristics.
100-200 mg orally twice daily for endometriosis; 200-400 mg twice daily for fibrocystic breast disease; 200 mg twice daily for hereditary angioedema. Maximum dose: 800 mg/day.

The terminal elimination half-life of testosterone after intramuscular injection of testosterone enanthate is approximately 8 days (range 4–12 days), reflecting slow absorption from the oily depot. This prolonged half-life supports a dosing interval of every 2–4 weeks.

Following intramuscular administration of testosterone enanthate, approximately 90% of the dose is excreted in urine as glucuronide and sulfate conjugates of testosterone and its metabolites (e.g., androsterone, etiocholanolone).
