Acetaminophen is primarily metabolized in the liver via conjugation with glucuronide (50-60%) and sulfate (20-30%). At toxic doses, glutathione is depleted, leading to NAPQI accumulation and hepatotoxicity.
Sulfamethoxazole is metabolized primarily via N-acetylation in the liver; phenazopyridine undergoes hepatic metabolism.
