XDEMVY (lotilaner ophthalmic solution) is a gamma-aminobutyric acid (GABA)-gated chloride channel antagonist. It inhibits the GABA-gated chloride channels in Demodex mites, leading to paralysis and death of the mites.
ACEPHEN (acetaminophen) is a para-aminophenol derivative with analgesic and antipyretic activity. Its mechanism involves inhibition of cyclooxygenase (COX) enzymes in the central nervous system, particularly COX-2, reducing prostaglandin synthesis. It has weak peripheral COX inhibition and minimal anti-inflammatory effect.

Acetaminophen is primarily metabolized in the liver via glucuronidation (UGT1A1, UGT1A6, UGT1A9) and sulfation (SULT1A1, SULT1A3). A minor fraction is oxidized by cytochrome P450 enzymes (CYP2E1, CYP1A2, CYP3A4) to a reactive toxic metabolite (NAPQI), which is normally detoxified by conjugation with glutathione.

Acetaminophen has been associated with cases of acute liver failure, at times resulting in liver transplant and death. Contact lenses should be removed prior to instillation and may be reinserted 15 minutes after dosing. Avoid touching the dropper tip to any surface to prevent contamination.
